CD47 (also known as the integrin-associated signal transducer) is a cell surface molecule in the immunoglobulin superfamily that has been reported to bind to several different proteins, including integrins, thrombospondin-1, and most importantly, signal regulatory protein a (SIRPa), which is a heavily glycosylated transmembrane protein with an immunoreceptor tyrosine-based inhibition motif (ITIM). CD47 occurs in all cells, including platelets. It is an unusual member of the Ig superfamily with a V-like extracellular domain, five membrane-spanning domains, and a short cytoplasmic domain. CD47 inhibition leads to stimulation of phagocytosis of cancer cells by macrophages. Anti-CD47 antibodies enhance the phagocytosis of tumor cells by macrophages by directly blocking the binding of tumor cell-expressing CD47 to macrophage-expressing SIRPa.Report Highlights
