Chromatography is a scientific discipline and a biophysical method used to separate, identify, and purify the components of a mixture for quantitative and qualitative analysis. This method works on the principle through which molecules of a mixture are coated onto a solid segment and components of the mixture are separated from each other via a stable segment, i.e., fluid stationary phase while working with the support of a mobile phase.
The most commonly used techniques in chromatography include gas chromatography (GC), high-performance liquid chromatography (HPLC), thin-layer chromatography, and paper chromatography, among which GC is the most common variant used.
Gas Chromatography (GC) is a method that enables segregating different types of compounds in a mixture by injecting a gaseous or liquid sample into a mobile phase and passing the carrier gas through a stationary phase. The mobile phase comprises an inert gas or an unreactive gas, such as helium, argon, nitrogen, or hydrogen, whereas the stationary phase is a solid or liquid that contains a separation column within the GC system. Mostly, GC columns are made up of fused silica capillaries with an inner diameter of 100-320 µm and a length of 5-60 µm, located inside an oven where the temperature of the gas is controlled and the effluent coming off the column is monitored by a suitable detector.
